Our poster will detail how the Pin-point platform was used to edit Hematopoietic Stem and Progenitor Cells (HSPCs). By optimizing reagent design and delivery conditions of a Pin-point base editor we achieved up to 80% C to T conversion at the target base in CD34+ HSPCs. Using the optimized conditions we targeted two separate loci known to reactivate γ-globin expression, which resulted in therapeutically relevant outcomes. Edited HSPCs retained viability, immunophenotype, and differentiation potential toward the erythroid lineage in vitro.
