SMN (Survival of motor neuron) is a 38-kD protein that is ubiquitously expressed in both the cytoplasm and nucleus. SMN plays an important role in assembling snRNP particles and is involved in RNA editing and transport. A lower expression of SMN has been correlated to the onset of Spinal Muscular Atrophy (SMA), an autosomal recessive disorder that is the leading hereditary cause of infant mortality, characterized by a loss of motoneurons in the spinal cord. The human SMN kit is designed for the detection of survival motor neuron protein directly in cell lysates.

Human SMN is measured using a sandwich immunoassay involving two specific human SMN antibodies, respectively labelled with Europium Cryptate (donor) and d2 (acceptor). The intensity of the signal is proportional to the concentration of SMN present in the sample.
The Human SMN assay features a two-plate assay protocol, where cells are plated, stimulated, and lysed in the same culture plate. Lysates are then transferred to the assay plate for the detection of SMN. This protocol enables the cells' viability and confluence to be monitored. The antibodies labelled with HTRF fluorophores may be pre-mixed and added in a single dispensing step to further streamline the assay procedure. The assay detection can be run in 96- to 384-well plates by simply resizing each addition volume proportionally.
