The AlphaLISA® immunoassay kit for AAV2, developed in cooperation with PROGEN, enables the quantitative determination of AAV2 capsids in buffer, cell culture media, cell lysates and cell supernatants using a homogeneous AlphaLISA assay (no wash steps).

  • Adeno-associated virus (AA) vectors are the leading platform for gene delivery for the treatment of a variety of human diseases. AAV Serotype 2 was one of the first AAV serotypes studied and used as vectors for specific and natural tropism towards targeted cell types such as neurons and hepatocytes or tissues such as skeletal muscles and the retina. The AAV2 kit is designed to detect and quantify AAV2 particles.

    AlphaLISA technology allows the detection of molecules of interest in a no-wash, highly sensitive, quantitative assay. In an AlphaLISA assay, a biotinylated anti-analyte antibody binds to the Streptavidin-coated Donor beads while another anti-analyte antibody is captured by AlphaLISA Acceptor beads. In the presence of the analyte, the beads come into close proximity. The excitation of the Donor beads causes the release of singlet oxygen molecules that triggers a cascade of energy transfer in the Acceptor beads, resulting in a sharp peak of light emission at 615 nm.
