Arrayed Screening

Arrayed screening offers a more targeted and detailed analysis of gene function. In this method, each gene perturbation is introduced into a separate well of a multi-well plate, enabling a clear correlation between the observed phenotype and the underlying genotype.

Arrayed screens are particularly advantageous for researchers interested in capturing detailed information on cellular responses to individual gene perturbations, why it is often used for confirmatory or validation screens. The primary read-out for arrayed screening is often imaging, which can be complemented with additional detection methods or sequencing techniques, facilitating a multi-parametric analysis. This approach proves especially useful for studying complex cell models, including primary cells and 3D cell cultures, where a higher level of experimental control is required.

In summary, arrayed screening provides researchers with a more comprehensive understanding of gene function by allowing the analysis of individual perturbations in a controlled environment. By leveraging imaging and other advanced analytical tools, scientists can explore the intricate relationship between genotype and phenotype, paving the way for enhanced drug discovery and precision medicine.

Advantages:

  • Multi-parametric read-outs
  • Suitable for more complex cell models
  • More choices in reagent formats, including non-viral transfection and cherry-picking options
  • Clear phenotype to genotype correlation
  • Improved editing efficiency by targeting the same gene with several gRNAs in one well

Read-out methods:

  • Fluorescence or luminescence detection to analyse Cell Viability and Proliferation
  • Imaging - Cell Morphology, Biomarker Expression and Localization
  • Live Cell Imaging – Differentiation, Migration
  • Proteomic and Metabolomic Profiling
