Guide
Revvity and DropGenie have collaborated to combine high-throughput automation and delicate biological precision into a user-friendly integrated solution, allowing scientists the opportunity to focus on biological insights rather than manual tasks.
The Revvity + DropGenie Solution
- Plug-and-Play Integration: The DropGenie platform is SBS-formatted, allowing the Revvity Fontus™ gripper to handle it like any standard plate.
- Precision Dispensing: Leverage the FlexDrop™ Plus for non-contact, nanoliter payload deposition to help minimize waste and increase experimental throughput.
- Move from initial setup to high-efficiency transfection in a new automation environment in less than five hours.

Why this automation matters
By miniaturizing the process, you can now run a 96-target screen in triplicate across four different donors using no more than 10 million cells per donor, extracting high-dimensional, multiomic data at a resolution previously reserved for small-scale pilot studies.
Versatility & Parallelization
- Payload flexibility: established protocols for mRNA, RNP (CRISPR), plasmid DNA, and Transposons.
- High parallelization: execute 48 independent reactions in parallel on a single microfluidic cartridge.
- Broad validation: in Primary T-cells (including T-regs), iPSCs, Synoviocytes, Myoblasts, and over 15 cell types.
triDrop, built for speed and gentle cell handling
The core of this technology is the Tri-Droplet Liquid Structure (triDrop). Unlike traditional systems that apply current directly to a bulk cell population in direct contact with the electrodes, our platform uses "liquid electrodes" to buffer cells from damage during electroporation.
- Thermal Protection: Cells are shielded from the heat and electrolysis byproducts (hydrolysis) that typically compromise viability.
- Immediate Recovery: Cells are automatically moved into a recovery buffer rapidly after the pulse.
- Scientific Result: Higher viability and faster recovery, supporting your downstream data reflects healthy biology, not experimental stress.
Researcher Advantage
The DropGenie+Revvity workflow provides a workhorse of discovery.
- Primary value: discovery and scale-out
- Cell input requirement: ultra-low (2-3% of standard)
- Workflow integration: plug-and-play (5-hr setup + calibration)
- Electroporation method: gentle "liquid electrode"
- Payload deposition: non-contact with the FlexDrop™ Plus dispenser
- Hardware flexibility: open SBS / standard deck
- Target use case: high-dimensional screens, cell engineering, arrayed CRISPR screening
- Workflow breadth: the Fontus™ workstation may also handle the complex upstream sample prep.

FAQs
-
How does this system compare to traditional "bulk" electroporation?
DropGenie enables you to achieve up to a 100x reduction in cell input and >85% CRISPR knockout in as few as 10,000 primary cells, compared to the millions required by conventional cuvette-based systems.
-
Can I use this with my existing Revvity protocol software?
Yes. The DropGenie module utilizes a REST API, making it natively compatible with the Revvity software environment.
-
Is it effective for CRISPR Knock-ins?
Absolutely. The platform is optimized for CRISPR-mediated knock-in, such as anti-HER2 CAR-T cell generation, requiring <1% of the cell input typically needed for these complex edits.
-
Does miniaturization affect the "Single Donor Resolution"?
It enhances it. Because the cell requirement is so low, you can perform extensive multiparametric assays on cells from a single donor, reducing the need to pool donors and mask individual biological variations.
