In the mouse, Interleukin 6 (IL-6) is a cytokine comprising 187 amino acids. In humans, IL-6 is produced by a variety of cell types, including T-cells, mast cells, and monocytes. IL-6 signals through binding to the gp130/ IL-6R receptor complex. IL-6 acts not only on the immune system, but also affects many physiological events in various organs. IL-6 exerts pro- or anti-inflammatory responses, depending on the target cell analyzed and the in vivo environmental circumstances. IL-6 is a differentiation and proliferation factor for B and T cells, and acts as a migration factor on monocytic cells. It is the major activator of acute-phase protein expression in the liver and acts as a survival factor on neuronal cells. Elevated serum IL-6 levels have been observed during bacterial and viral infection, trauma, autoimmune disease, inflammation, and cancer.

AlphaLISA technology allows the detection of molecules of interest in a no-wash, highly sensitive, quantitative assay. In an AlphaLISA assay, a biotinylated anti-analyte antibody binds to the Streptavidin-coated Donor beads while another anti-analyte antibody is conjugated to AlphaLISA Acceptor beads. In the presence of the analyte, the beads come into close proximity. The excitation of the Donor beads causes the release of singlet oxygen molecules that triggers a cascade of energy transfer in the Acceptor beads, resulting in a sharp peak of light emission at 615 nm.
