Myosins are actin-based motor proteins that contribute to the generation of contractile force in eukaryotic cells. Each muscle myosin assembly contains a pair of myosin heavy chain (MYH) and two pairs of myosin light chain (MYL). MyHC-2 is a member of the class II or conventional myosin heavy chains, and functions in skeletal muscle contraction. This assay enables the detection of Myosin Heavy Chain in cell lysates for research related to skeletal muscle diseases.

Myosin Heavy Chain is measured using a sandwich immunoassay involving two specific anti-Myosin Heavy Chain antibodies, respectively labelled with Europium Cryptate (donor) and d2 (acceptor). The intensity of the signal is proportional to the concentration of the Myosin Heavy Chain present in the sample
The Myosin Heavy Chain assay protocol is described here. Cells are plated, stimulated, and lysed in the same 96-well culture plate. Lysates are then transferred to the assay plate for the detection of Myosin Heavy Chain. This protocol enables the cells' viability and confluence to be monitored. The antibodies labelled with HTRF fluorophores may be pre-mixed and added in a single dispensing step to further streamline the assay procedure. The assay detection can be run in 96- to 384-well plates by simply resizing each addition volume proportionally.
C2C12 cells were grown in presence of different percentages of Fetal Bovine Serum (FBS) or Horse Serum (FS) to induce differentiation of C2C12 myoblasts into myotubes. After 4- or 8-day incubation, media was aspirated and cells were lysed with lysis buffer 1X for 30 min at RT under gentle shaking. 16µL of lysate were transferred into a 384-well sv white microplate and 4 µL of the Myosin Heavy Chain detection reagents were added. The HTRF signal was recorded after an overnight incubation at RT.
