Overview

Our ready-to-use PhenoVue DNA damage staining kit offers a reliable and straightforward solution for visualizing key markers of DNA damage and repair:

  • γ-H2AX (Phospho S139 H2AX) - an early marker of double-strand breaks (DSBs), detected as distinct nuclear foci.
  • 53BP1 - recruited to DSB sites and involved in non-homologous end joining (NHEJ) repair, forming punctate foci.
  • Nucleus morphology - analyzed using PhenoLOGIC™ (in Harmony™ image analysis software) to assess late apoptosis through cell shrinkage and DNA fragmentation.

For enhanced flexibility, up to two additional biological markers such as PARP, ATM, or CHK can be included using the PhenoVue Fluor 555 and 400LS channels, allowing for deeper insights into DNA damage response (DDR) mechanisms.

The PhenoVue DNA Damage Staining Kit supports multiple drug discovery applications including:

  • Oncology - allows the study of DDR-targeting therapeutics (e.g. PARP, ATM, ATR inhibitors), identification of synthetic lethal interactions (e.g. BRCA mutations), and assessment of genotoxic and cytotoxic effects.
