AlphaLISA technology allows the detection of molecules of interest in buffer, cell culture media, serum and plasma in a highly sensitive, quantitative, reproducible and user-friendly mode. In an AlphaLISA assay, a Biotinylated Anti-Analyte Antibody binds to the Streptavidin-coated Alpha Donor beads, while another Anti-Analyte Antibody is conjugated to AlphaLISA Acceptor beads. In the presence of the analyte, the beads come into close proximity. The excitation of the Donor beads provokes the release of singlet oxygen molecules that triggers a cascade of energy transfer in the Acceptor beads, resulting in a sharp peak of light emission at 615 nm.
TNF-related apoptosis-inducing ligand (TRAIL) is a type II transmembrane protein expressed on the cell surface that is also known as tumor necrosis factor superfamily 10 (TNFSF10). This transmembrane protein can be cleaved by cysteine proteinases at the surface, which in turn generates the soluble form of TRAIL. TRAIL has a molecular weight of approximately 24-28 kDa and functions as a ligand, inducing the process of cell death primarily in tumor cells by binding to certain death receptors. TRAIL and its receptors are currently used for several anti-cancer therapeutics and is involved in the pathophysiological processes of cardiovascular and metabolic diseases.
