The AlphaLISA™ immunoassay kit for human MMP-8 enables the detection and quantitation of human MMP-8 in human serum or cell culture medium using a homogeneous AlphaLISA assay (no wash steps). The kit was designed to detect both MMP8 and pro-MMP8.

Matrix metalloproteinase 8 (MMP8, Neutrophil collagenase or PMNL collagenase), a member of the matrix metalloproteinase family, is an enzyme composed of a pro-domain that is cleaved off for activation. MMP 8 is involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. Increased serum levels of MMP8 have observed to be a strong predictor of mortality in patients with Rheumatoid Arthritis. Further, MMP8 serum levels show evidence of being a valuable biomarker for cancers such as colorectal cancer. This kit has been designed for the detection of Human MMP 8 in serum and cell culture supernatants.
AlphaLISA technology allows the detection of molecules of interest in a no-wash, highly sensitive, quantitative assay. In an AlphaLISA assay, a biotinylated anti-analyte antibody binds to the Streptavidin-coated Donor beads while another anti-analyte antibody is conjugated to AlphaLISA Acceptor beads. In the presence of the analyte, the beads come into close proximity. The excitation of the Donor beads causes the release of singlet oxygen molecules that triggers a cascade of energy transfer in the Acceptor beads, resulting in a sharp peak of light emission at 615 nm.
