Measurements of hAFP and the Free hCGß subunit of hCG from maternal serum serve as an aid in diagnosing fetal malformations, e.g. neural tube defects and Down´s syndrome. These solid-phase, two-site fluoroimmunometric assays are based on the direct sandwich technique in which hAFP is detected using two monoclonal antibodies directed against two seperate separate antigenic determinants on the hAFP molecule. Free hCGß is detected in a similar way using two different monoclonal anti-hCGß antibodies.
