IVISense 750 fluorescent dyes are available in self-quenching format as NHS esters or self-quenching maleimide reactive dyes for conjugation to either free amine (-NH3) or free thiol (-SH) containing molecules.

Overview

IVISense 750 NHS fluorescent dye is an amine-reactive near-infrared (NIR) fluorochrome for coupling via an N-Hydroxysuccinimide (NHS)-ester linkage to peptides, small molecules, proteins, antibodies or macromolecules. Unit size 1 mg.

IVISense™ 750 fluorescent dyes are ideal for molecular labeling for use in preclinical in vivo imaging applications due to enhanced tissue penetration at this wavelength. IVISense 750 dyes are well suited for the labeling and imaging of proteins, antibodies, and small molecules in a wide range of research areas. The 750 nm fluorescence wavelength is ideal for biological imaging applications using in vivo fluorescence systems but will only work on microscopes, high content imaging and flow cytometry systems that have appropriate light sources and filter sets to accommodate this wavelength.

  • Uniquely optimized for the generation of in vivo imaging probes with low non-specific accumulation at tumor sites and inflammatory sites.
  • Need special microscopes, flow cytometers, and plate readers that accommodate excitation and emission with light sources and filters for this wavelength.
  • Suitable for in vivo multiplexing with 645 or 680 nm Revvity probes or other labeled proteins/Abs.  Expanded in vivo multiplexing with IVIS spectral unmixing with 645/680/800 nm probes or antibodies.
  • No spectral interference with mouse diets, so alfalfa-containing mouse diets can be used.
  • Recommended dye:protein ratio of 1-3.

IVISense 680 NHS 673/691 When a protein is labeled with multiple IVISense Self-Quenching Dye molecules, there is a shift in the absorbance of the dye, which will reduce the signal.

Only choose IVISense Self-Quenching Dyes if your labeling ratio of dye:protein will be 1:1. If multiple dyes are to be conjugated onto your protein, a non-self-quenching IVISense Dye should be used. 

NHS and MAL dyes are suitable as noted above.
