Endogenous detection of toll-like receptor-mediated IRF5 signaling using AlphaLISA SureFire® Ultra

Interferon regulatory factor 5 (IRF5) is a master regulator of innate immunity, driving the expression of type I interferons and pro-inflammatory cytokines in response to Toll-like receptor (TLR) activation. Dysregulation of IRF5 signaling has been linked to autoimmune diseases and cancer, making it a critical target for immunology research.

This application note demonstrates how AlphaLISA SureFire® Ultra™ technology  allows for no-wash detection of endogenous IRF5 activation—including phosphorylation (Ser446) and dimerization—in human immune cells. The assay also profiles upstream signaling events involving TAK1, IKKα/β, and NFκB p65, providing a complete view of IRF5 pathway activation.

Key highlights include:

  • Robust detection of total, phosphorylated, and dimerized IRF5
  • Profiling of TLR7/8/9-induced signaling cascades
  • Optimization guidance for low cell input assays
  • Multiplexable, no-wash workflow for high-throughput studies

Discover how AlphaLISA SureFire® Ultra assays can elevate your IRF5 research and accelerate pathway-focused discovery.
