Optimizing nanobody-engineered AAV manufacturing for scalable precision gene delivery

AAV-Nanobody manufacturing optimization through design of experiments and functional analysis

As targeted AAV vectors become increasingly sophisticated, improving manufacturability without compromising functionality remains a key challenge.

For AAV-nanobody constructs, production parameters can directly influence both vector yields and the number of nanobodies displayed on each capsid, affecting downstream performance.

In this ASGCT 2026 poster, Revvity scientists applied a Design of Experiments (DoE) approach to optimize AAV9-nanobody manufacturing and identify conditions that improve productivity while maintaining vector functionality.

The study demonstrates how plasmid ratio optimization can increase vector yields, fine-tune nanobody incorporation, and support more efficient production of targeted AAV vectors.
