The SARS-CoV-2 Spike S1 protein is one of two part of the virus Spike protein (S). The S1 subunit is responsible for the virus interaction with host cells' transmembrane Angiotensin-converting enzyme 2 receptors (ACE2), while the S2 subunit is capable of conformational changes that allow the fusion of the viral particle envelope and host cell membrane upon S1/ACE2 binding. As such, the S1 protein is closely investigated for its role in viral infection and is a reliable marker of infection as well as a potential vaccine immunogen target.

AlphaLISA technology allows the detection of molecules of interest in a no-wash, highly sensitive, quantitative assay. In an AlphaLISA assay, a biotinylated anti-analyte antibody binds to the Streptavidin-coated Donor beads while another anti-analyte antibody is conjugated to AlphaLISA Acceptor beads. In the presence of the analyte, the beads come into close proximity. The excitation of the Donor beads causes the release of singlet oxygen molecules that triggers a cascade of energy transfer in the Acceptor beads, resulting in a sharp peak of light emission at 615 nm.
