Human immune system multiomics studies: from high dimensional immunosurveillance to targeted hypothesis follow up

A presentation by Julian Reading, Senior Manager, Flow Cytometry at the Allen Institute for Immunology.

The Allen Institute for Immunology recently completed a 3-year longitudinal multiomics study with cohorts from multiple autoimmune and oncology clinical centers. After developing high dimensional, highly robust protocols for immune cell interrogation by flow cytometry and single cell genomics we now face a new challenge. Generating smaller, targeted, panels to follow up on patterns and novel hypotheses revealed following data analysis. The successfully integrated cell counting and cell sorting components in phase 1, have been reimagined for high throughput, lower dimensional studies in phase 2. In this presentation I will highlight our integration of technologies with automation systems, which systems are retained across both approaches to human immune studies and where, in our need to scale up and automate, we have made substitutions. These approaches and pivots are common in modern medicine development, and I hope that the work we are doing will have broad appeal to the scientific community at large.
